چکیده

The theory of light scattering indicates that aggregation of the crystalline lens proteins can produce opacity. Furthermore, it is known that calcium increase? in the cataractous lens. We have, therefore, investigated the effectiveness of calcium ion in producmg aggregates of the soluble proteins of the bovine lens. The experimental restdts show: (1) CaCU produce's aggregates of the soluble protein of the lens; (2) the pre-a and at fraction of the lens protein are the specific targets of calcium ion even in the presence of sidfhydryl reducing reagent; (3) the aggregation can be reversed and transparency restored when calcium is removed either by dialysis or chelation; and (4) the molecular weight distribution and index of refraction of the aggregates have been measured.

Comparison of ultraviolet induced photo-kinetics for lens-derived and recombinant beta-crystallins.

PURPOSE The photobiology of purified recombinant crystallins has not been studied. Here we examine photo-induced aggregation of purified recombinant mouse betaA3-crystallin (rbetaA3) and compare it with that of betaL-crystallins isolated from bovine lenses. METHODS rbetaA3-Crystallin was expressed in baculovirus-infected Sf9 cells and purified by ion-exchange and gel-filtration chromatography...

Alpha-crystallin can function as a molecular chaperone.

The alpha-crystallins (alpha A and alpha B) are major lens structural proteins of the vertebrate eye that are related to the small heat shock protein family. In addition, crystallins (especially alpha B) are found in many cells and organs outside the lens, and alpha B is overexpressed in several neurological disorders and in cell lines under stress conditions. Here I show that alpha-crystallin ...

Age-dependent association of γ-crystallins with aged α-crystallins from old bovine lens

PURPOSE Previous theoretical and experimental studies have predicted that the loss of weak protein interactions between alpha- and gamma-crystallins could result in a decrease in the transparent properties of the aging lens. METHODS alpha-Crystallins were prepared from the nucleus of old bovine lens, and gamma-crystallins were prepared from whole fetal bovine lens or from the nucleus of old b...

Aggregation in Aqueous Solutions of Bovine Lens y-Crystallins: Special Role of ys

RESULTS. All of the y-crystallins studied formed large aggregates (or "megamers") in aqueous solutions. However, each protein differed in the relative rates of formation of megamers. ysCrystallin formed megamers much more slowly than y,,and yIVa-crystallin. In solutions containing mixtures of y,, and ys, and y,Va and ys, ys-crystallin significantly suppressed the aggregation of y,,and yIV:i-cry...

Effect of dicarbonyl-induced browning on alpha-crystallin chaperone-like activity: physiological significance and caveats of in vitro aggregation assays.

Alpha-crystallin is a member of the small heat-shock protein family and functions like a molecular chaperone, and may thus help in maintaining the transparency of the eye lens by protecting the lens proteins from various stress conditions. Non-enzymic glycation of long-lived proteins has been implicated in several age- and diabetes-related complications, including cataract. Dicarbonyl compounds...
